Avant de devenir religieuse, Virdiana sort une dernière fois du couvent pour se rendre chez son riche oncle Don Jaime. Celui-ci habite seul dans son domaine depuis la mort de sa femme, et est submergé par la beauté de Viridiana, qu'il demande en mariage. La jeune femme tente alors de s'enfuir, apeurée par son insistance, lorsqu'elle apprend que Don Jaime s'est suicidé par désespoir. Héritant du domaine, Viridiana décide de ne pas rentrer dans les ordres et de se consacrer aux mendiants des environs. Peu à peu, la jeune femme se retrouve sous leur coupe...
