1935, dans un cabaret de Shangaï, l’archéologue Indiana Jones négocie avec de redoutables Chinois la possession d’un joyau exceptionnel, doté de pouvoirs magiques. Lorsque les négociations tournent mal, Jones emmène à sa suite la chanteuse Willie Scott, elle aussi intéressée par la pierre précieuse, et Demi-Lune, un jeune Chinois débrouillard. La course folle se termine au cœur de la jungle, en Inde. Les trois acolytes y découvrent un temple souterrain où sévit une secte dirigée par le mollah Ram. Cette secte, qui se livre régulièrement à des sacrifices humains, a réduit en esclavage tous les enfants de la région, et les oblige à travailler dans la mine. De plus, le mollah Ram a réunit trois joyaux, dont celui tant convoité par Indiana Jones. N’écoutant que son courage, le plus célèbre des aventuriers décide d’agir et vole au secours des victimes.
